The large amount of molasses that are generated in sugar-processing companies are not always redistributed for commercialization in by-products. Because of this, the present research uses these wastes as fuel in low-cost, lab-scale, single-chamber microbial fuel cells. Zinc and copper electrodes were used as electrodes and 100 mL of molasse in the chamber as fuel, managing to generate current and voltage peaks of 1.73 ± 0.13 mA and 0.953 ± 0.142 V. In monitoring the conductivity of the substrate, a maximum peak of 111.156 ± 8.45 mS/cm was observed, and a slightly acidic pH was observed throughout the monitoring. It was possible to obtain a power density of 5.45 ± 0.31 W/cm2 for a current density of 308.06 mA/cm2, while the yeast count showed a logarithmic curve throughout the monitoring. Finally, the molecular technique identified 100% of the special C. boidinii present in the anodic...

Background: Cell-free DNA (cfDNA) is used in clinical research to identify biomarkers for diagnosis of and follow-up on cancer. Here, we propose a fast and innovative approach using traditional housekeeping genes as cfDNA targets in a copy number analysis. We focus on the application of highly sensitive technology such as digital PCR (dPCR) to differentiate breast cancer (BC) patients and controls by quantifying regions of PUM1 and RPPH1 (RNase P) in plasma samples. Methods: We conducted a case-control study with 82 BC patients and 82 healthy women. cfDNA was isolated from plasma using magnetic beads and quantified by spectrophotometry to estimate total cfDNA. Then, both PUM1 and RPPH1 genes were specifically quantified by dPCR. Data analysis was calibrated using a reference genomic DNA in different concentrations. Results: We found RNase P and PUM1 values were correlated in the patient g...

Analysis of immune responses in Bartonella bacilliformis carriers are needed to understand acquisition of immunity to Carrion’s disease and may allow identifying biomarkers associated with bacterial infection and disease phases. Serum samples from 144 healthy subjects from 5 villages in the North of Peru collected in 2014 were analyzed. Four villages had a Carrion’s disease outbreak in 2013, and the other is a traditionally endemic area. Thirty cytokines, chemokines and growth factors were determined in sera by fluorescent bead-based quantitative suspension array technology, and analyzed in relation to available data on bacteremia quantified by RT-PCR, and IgM and IgG levels measured by ELISA against B. bacilliformis lysates. The presence of bacteremia was associated with low concentrations of HGF (p = 0.005), IL-15 (p = 0.002), IL-6 (p = 0.05), IP-10 (p = 0.008), MIG (p = 0.03) and ...

Acidophilic microorganisms are predominant in the ecosystems of acid mine waters, being found in abundance the species Acidithiobacillus ferrooxidans. The objective of the study was to produce electricity in the Microbial Fuel Cells (MFC) from thiosulfate at the anode and ferric ion at the cathode using the bacteria At. ferrooxidans LB151, which was characterized morphologically and molecularly. The electrochemical techniques used were chronoamperometry and cyclic voltammetry. The cyclic voltammetry show current peaks of oxidation and reduction of the compounds indicating that there is an interaction of the bacteria with the surface of the electrode. The voltage generated by the MFC is greater than the control cell, producing an average power density of 24.23 mW/m2 in 4 hours while the control cell produced an average power density of 3.41 mW/m2 in the same time. The total energy generat...

Low- and middle-income countries (LMICs) are significantly affected by SARS-CoV-2, partially due to their limited capacity for local production and implementation of molecular testing. Here, we provide detailed methods and validation of a molecular toolkit that can be readily produced and deployed using laboratory equipment available in LMICs. Our results show that lab-scale production of enzymes and nucleic acids can supply over 50,000 tests per production batch. The optimized one-step RT-PCR coupled to CRISPR-Cas12a-mediated detection showed a limit of detection of 102 ge/μL in a turnaround time of 2 h. The clinical validation indicated an overall sensitivity of 80%–88%, while for middle and high viral load samples (Cq ≤ 31) the sensitivity was 92%–100%. The specificity was 96%–100% regardless of viral load. Furthermore, we show that the toolkit can be used with the mobile lab...
