The cryoprotectant effect of two hypertonic extenders (trehalose and lactose) on the post-thawing characteristics of ram semen (n=4) was evaluated. The extender composition included Tris 27.1 g/l, Citric acid 14.0 g/l, Fructose 10.0 g/l, Glycine 10.0 g/l, egg yolk 10.0% (v/v) and Glycerol 6.5% (v/v). Semen was collected in an artificial vagina. Seminal characteristics were: volume: 1.1 ± 0.1 ml, sperm concentration: 3.50 ± 0.1 x 109/ml, individual motility: 87.0 ± 2.4%, wave motility (scale 0-5): 4.4 ± 0.2, live sperms: 90.2 ± 3.8%, and abnormal sperms: 1.8 ± 0.7%. Semen was frozen in 0.5 ml straws and stored in liquid nitrogen. Straws were thawed after 3 months. Extenders have been established as an alternative to compensate the antioxidant deficiencies of stallion semen during freezing. Several research works have aimed to evaluate the effect of various additives with antioxidant properties on the equine sperm cryotolerance; however, in limited occasions this has been linked with the extender total antioxidant capacity (TAC). The objective of the study was to evaluate the effects of two semen extenders: Tris- Fructose-egg yolk (Tris) and Citrate-Glucose-egg yolk (citrate) on motility and sperm membrane integrity (HOST) in ovine frozen semen in pellets. The study was carried out at the Semen Bank of the Agrarian University La Molina, in Lima, Peru, using 4 rams (2 Assaf and 2 Blackbelly) of 3.5 to 4 years old. A covariance analysis was used to evaluate the effect of the treatment and breed on Individual Progressive Motility (IPM), and randomized block design to evaluate the effect of extenders on sperm membrane integrity. The aim of this study was to evaluate sodium caseinate (CS) in the dilution and freezing of bovine semen. Fifteen ejaculates from five bulls were diluted and then individually frozen using a diluent composed of tris-glycerol-citric acid with 8% v/v of low-density lipoproteins (LDL) and supplemented with CS in four treatments: CS 3% w/v (CS3), CS 4% w/v (CS4), CS 5% w/v (CS5) and without CS (Control). The effect of two semen extenders: Glucose Tris and Ovine Freezing Buffer (UA 466/ 005238) on the motility and cytoplasmic membrane integrity of spermatozoa during the freezing process was evaluated. Five rams (2 Assaf, 2 Cinnamon and 1 Black Belly) were used. The fresh semen was of good quality and values of seminal characteristics were within the normal range for this species. Se evaluó el efecto del contenido de materia grasa (%MG), en leche descremada UHT usada como diluyente seminal sobre la integridad de membrana plasmática y la motilidad progresiva (MP) de espermatozoides caninos preservados a 4 °C. Se obtuvieron 20 eyaculados mediante manipulación digital de 10 perros Bulldog Inglés y American Bully de fertilidad probada. Cada eyaculado fue evaluado a través de espermiograma convencional y de una prueba hipoosmótica simplificada (HOST-s). Las muestras fueron fraccionadas y diluidas en relación 1:3 con leche descremada UHT como diluyente a 0%MG y 0.5%MG, con adición de estreptomicina (50 mg/100 ml) y penicilina sódica (50 mg/100 ml). The aim of this study was to evaluate the post-thawing quality of equine semen subjected to two dimethylformamide (DMF) addition schemes during freezing. El objetivo de esta investigación fue evaluar el uso de yema de huevo centrifugada (YHC) en la congelación de espermatozoides epididimales caninos. Se obtuvieron los complejos testículo-epidídimo (CTE) después de la orquiectomía de 20 perros domésticos (Canis familiaris) de varias razas y en un rango de edad entre 1 y 6 años. The effect of glucose, fructose, trehalose, and sucrose, as components of the extender, was evaluated on canine sperm viability during refrigeration at 5 ºC. The spermatic fraction of 16 dog ejaculations were distributed in 5 aliquots containing an extender based on Tris – citric acid – egg yolk, but differing in the presence of one of the sugars, plus a control without sugar. The five treatments were evaluated after 24, 48, 72, and 96 hours of refrigeration in relation to progressive motility and functional integrity of the membrane by the hypoosmotic swelling test (HOS). All groups, with the exception of the control group, the sperm motility maintained within acceptable ranges during the study (90% on day 0 and 60% on day 4). Thirty-two semen samples from four rams were frozed using two antioxidants, Tempo (2,2,6,6 tetramethyl-1-piperidinyloxyl) and Tempol (4-hidroxi 2,2,6,6 tetramethyl-1- piperidinyloxyl), each one in concentrations of 0.5, 1.0, and 2.5 mM, to evaluate the effects on post-thawing motility, viability, acrosomal integrity, and earlier sperm capacitation. Eight replications for each antioxidant concentration and control group, were done using four semen samples per replication. Semen samples were diluted on a Tris extender. Results showed that Tempo 0.5 mM improved frozen semen quality in comparison with control group (p<0.05), by increasing progresive motility (79 vs. 67%), viability and acrosomal integrity (70 vs. 58%), and decreasing earlier sperm capacitation (9 vs. 15%). Se analizó el efecto de los dilutores Tris-glucosa y Ovine Freezing Buffer (UA 466/ 005238) sobre la motilidad e integridad de la membrana citoplasmática de los espermatozoides durante el proceso de congelación de semen ovino. Se utilizó el semen de cinco carneros (2 Assaf, 2 Canela y 1 Black Belly). El semen fresco fue de buena calidad y los valores de las características seminales estuvieron dentro de los parámetros de la especie.
