The aim of this study was to characterize the PepT1 of pirarucu Arapaima gigas through genomic and proteomic tools. A transcriptomic and proteomic analysis was made from sections of the intestine, spleen, liver and kidney of fingerlings. The transcriptomic analysis allowed obtaining two consensus sequences of 357 and 459 bp. Both fragments showed a high degree of homology (78 and 80%), mainly with nucleotide sequences coding for the PepT1 of Scleropages formosus of the same Order as A. gigas. The proteomic analysis by double mass spectrometry MALDI TOF/TOF allowed to identify 15 peptide sequences of PepT1 in pirarucu. In conclusion, PepT1 was partially characterized in the intestine of pirarucu, which could be used for further studies on the evaluation of its expression.

The chame Dormitator latifrons is a symbolic species associated with rural aquaculture in coastal regions of the South American Pacific. Its adaptation to stay alive out of water (anhydrobiosis) for days is beneficial for its storage and transport at room temperature. The aim of the study was to determine the tolerance of chame to anhydrous conditions. Sixty chames were distributed in containers and kept in two climate-controlled environments at 16 and 24 °C for 96 h. Survival, malondialdehyde levels, total proteins and pH in the muscles and lactate dehydrogenase (LDH) activity in the gills were monitored. Additionally, the behaviour of the fish was monitored, as well as the total number of microorganisms with a sensory evaluation. The critical period of survival occurred after 48 h, where the pH decreased, and the MDA levels increased. The number of microorganisms increased in both en...

The aim of this study was to evaluate the sedative effect of three products: valerian extract (Valeriana officinalis), benzocaine and tricaine methanesulfonate (MS-222) on stress levels produced by a simulated transport model in tilapia fingerlings. In a first part, the sedative dose of the products was determined based on the clinical characteristics that indicate depression of the central nervous system. In the second part, sedation and subsequent simulated transport for 3 hours were performed. The fish were euthanized, and blood samples were taken from the tail vein to determine cortisol (ELISA) and glucose (portable glucometer) levels, haematological parameters and mortality. The optimal doses of sedation were 200, 35 and 75 mg/l, respectively, for valerian extract, benzocaine and MS-222. After 3 h of simulated transport, cortisol and glucose values were similar or higher compared to...

The aim of this study was to determine the postnatal development of Islets of Langerhans (IL) in alpacas of 0 to 45 days of age. The animals were divided in nine age groups with range of five days. The mumber of IL varied from 3.03 to 4.46 in 10x fields (with an ocular of 10x), the IL diameter varied from 67.89 to 72.55 μm, the number of β cells in the IL from 23.79 to 47.08 and other cells from 18.15 to 39.93. Moreover, the insulin receptor and the transport protein GLUT-2 were observed by immunohistochemistry on the first day of life. It is concluded that the number and size of IL increased with age (p<0.05) while β cells do not show the same trend.

Iron is essential for health but in excess may be harmful. The living organism has ability to store and re-use the iron body and as such its requirement is small (1-2 mg absorbed/day). Hepcidin, a hormone produced in the liver, has an important role in iron homeostasis by blocking the iron transport protein and inhibiting iron absorption. Maternal anemia due to iron deficiency is a public health problem when it is moderate (7-<9 g/dL) and severe (Hb<7 g/dL) since it increases the risk of maternal and neonatal morbi-mortality. Similarly, high levels of hemoglobin during pregnancy affect the mother and newborn. In Peru, there is a National Program to supplement with iron to all pregnant women anemic or not. Scientific evidence shows that hemoglobin values between 9-10.5 g/dL are optimal for best fetal growth and that treatment with iron supplementation to non anemic women increases levels ...

Potato virus X (PVX) occurs worldwide and causes an important potato disease. Complete PVX genomes were obtained from 326 new isolates from Peru, which is within the potato crop′s main domestication center, 10 from historical PVX isolates from the Andes (Bolivia, Peru) or Europe (UK), and three from Africa (Burundi). Concatenated open reading frames (ORFs) from these genomes plus 49 published genomic sequences were analyzed. Only 18 of them were recombinants, 17 of them Peruvian. A phylogeny of the non-recombinant sequences found two major (I, II) and five minor (I-1, I-2, II-1, II-2, II-3) phylogroups, which included 12 statistically supported clusters. Analysis of 488 coat protein (CP) gene sequences, including 128 published previously, gave a completely congruent phylogeny. Among the minor phylogroups, I-2 and II-3 only contained Andean isolates, I-1 and II-2 were of both Andean and...

The empirical use of alpaca rennet in the production of artisanal cheeses with cow’s milk is widespread in the high Andean communities of Perú and Bolivia, and has not been scientifically reported. The objective of the present study was to characterize alpaca rennet and evaluate its coagulating activity. Ready-to-use alpaca curds were collected directly from dairies in four neighborhoods of Viscapalca (Huancavelica-Perú, 4003 masl), which were transported under refrigeration and kept frozen at -20°C. The Crude Enzymatic Extract (CEE) was obtained by centrifugation at 560 x g for 10 min at 4°C, from which proteins were determined by Bradford, SDS-PAGE electrophoretic profile, coagulation time and proteolytic activity. The traditional management of alpaca rennet is also described. CEE was found to contain 29.80 to 52.91 µg/ml of protein. The SDS-PAGE electrophoretic profile revealed...
