The development of new strategies based on the use of Tr1 cells has taken relevance to induce long-term tolerance, especially in the context of allogeneic stem cell transplantation. Although Tr1 cells are currently identified by the coexpression of CD49b and LAG-3 and high production of interleukin 10 (IL-10), recent studies have shown the need for a more exhaustive characterization, including co-inhibitory and chemokines receptors expression, to ensure bona fide Tr1 cells to be used as cell therapy in solid organ transplantation. Moreover, the proinflammatory environment induced by the allograft could affect the suppressive function of Treg cells, therefore stability of Tr1 cells needs to be further investigated. Here, we establish a new protocol that allows long-term in vitro expansion of highly purified expanded allospecific Tr1 (Exp-allo Tr1). Our expanded Tr1 cell population becomes...

Some dye-sensitized solar cells were prepared using TiO2 modified with CuO (TiO2-CuO) like a working electrode. P25 commercial powder and small amounts of CuO powder were used in preparing the modified coating. The coating was deposited following doctor Blade method, on a conductive substrate SnO2:F, which was previously deposited on a TiO2 blocking layer by spray pyrolysis technique. For sensitization of the coating, a ruthenium complex, cis-bis (isothiocyanate) bis (2,2'bipiridil-4,4'dicarboxilato) ruthenium (II) ammonium bistetrabutil, also known as N719, was used. The CuO powder was structurally characterized by X-ray diffraction and its morphology was studied by scanning electron microscopy. The functional groups of TiO2 and CuO were studied by infrared spectroscopy. The oxidation and reduction cycles of Cu were shown by cyclic voltammetry. Through incident photon to current convers...

“Cotton is the most widely used natural fiber for textiles; however, the capacity of cotton fibers to absorb large amounts of moisture, retain oxygen, and have a high specific surface area makes them more prone to microbial contamination, becoming an appropriate medium for the growth of bacteria and fungi. In recent years, the incorporation of silver nanoparticles in textile products has been widely used due to their broad-spectrum antibacterial activity and low toxicity towards mammalian cells. The aim of the current study is to continue the assessment of our developed nanocomposite and evaluate the antibacterial and antifungal activity of the nanocomposite based on silver nanoparticles and carboxymethyl chitosan (AgNPs-CMC) against Escherichia coli, Staphylococcus aureus, and Candida albicans, evaluated by the well diffusion method. The antibacterial activity against E. coli and S. a...

ackground: Depression and anxiety are two of the most prevalent and disabling mental disorders worldwide, both in the general population and in outpatient clinical settings. Objective: This study aimed to analyze the psychometric properties of the Patient Health Questionnaire-4 (PHQ-4) based on network analysis metrics. Methods: A total of 911 Paraguayans (23.71% women and 76.29% men; mean age 31.25 years, SD = 10.63), selected by non-probabilistic convenience sampling, participated in the study. Network analysis was used to evaluate the internal structure, reliability, and measurement invariance between men and women. Results: The results revealed that the PHQ-4 is a unidimensional measure through Exploratory Graph Analysis (EGA). Reliability, through structural consistency, identified that 100% of the time, only a single dimension was obtained, and all items remained stable, as they we...

This work studies the interaction between natural dyes obtained from Peruvian Zea mays and Bixa orellana seeds and nanostructured titanium dioxide, in order to evaluate their function as sensitizers into solar cell devices. The effective attachment of dyes to the TiO2 layer is corroborated by the comparison of UV-Visible absorption and FT-IR spectra of the extracted dye solutions and sensitized TiO2 electrodes. The principal compounds from the seed extraction of Zea mays and Bixa orellana are cyanidin-3-glucoside (C3G) and bixin respectively, which were analyzed in an isolated dye/cluster TiO2 system by molecular dynamic simulation. The results showed that chemisorption is carried out through a consecutive deprotonation process, and Ti-O bond formation by the monodentate OH and COOH anchoring groups, for C3G and bixin respectively. Finally, we tested the effect of the dye - TiO2 interact...

Ischemic brain injury represents a major cause of death worldwide with limited treatment options with a narrow therapeutic window. Accordingly, novel treatments that extend the treatment from the early neuroprotective stage to the late regenerative phase may accommodate a much larger number of stroke patients. To this end, stem cell-based regenerative therapies may address this unmet clinical need. Several stem cell therapies have been tested as potentially exhibiting the capacity to regenerate the stroke brain. Based on the long track record and safety profile of transplantable stem cells for hematologic diseases, bone marrow-derived mesenchymal stromal cells or mesenchymal stromal cells have been widely tested in stroke animal models and have reached clinical trials. However, despite the translational promise of MSCs, probing cell function remains to be fully elucidated. Recognizing th...

In this paper a hybrid scheme given by diffusion equation in conjunction to classical electrodynamics to examine physics-based mechanisms behind the behavior of beta cells, is presented. Particular attention is paid on the scenario by which beta-cell is not able to segregate insulin granules. Thus, a mechanism that would explain anomalies as seen in the lost of functions to segregate granules of insulin, is proposed. In this manner, the main argument of this paper states that the processes previous to the segregation of insulin given by (i) entrance of insulin, (ii) exit of Potassium K+, (iii) entrance of Calcium 2+, and (iv) segregation of insulin belong in only one universal formulation. A pseudo code by which the diabetes disease might be seen as a bug of the biological software of beta cell is proposed. Therefore a scheme capable to fix that bug would constitute a path to alleviate t...

The protozoan parasite Leishmania (Viannia) braziliensis (L. braziliensis) is the main cause of human tegumentary leishmaniasis in the New World, a disease affecting the skin and/or mucosal tissues. Despite its importance, the study of the unique biology of L. braziliensis through reverse genetics analyses has so far lagged behind in comparison with Old World Leishmania spp. In this study, we successfully applied a cloning-free, PCR-based CRISPR–Cas9 technology in L. braziliensis that was previously developed for Old World Leishmania major and New World L. mexicana species. As proof of principle, we demonstrate the targeted replacement of a transgene (eGFP) and two L. braziliensis single-copy genes (HSP23 and HSP100). We obtained homozygous Cas9-free HSP23-and HSP100-null mutants in L. braziliensis that matched the phenotypes reported previously for the respective L. donovani null muta...

Paneth cells have an important function in the defense and protection mechanisms of the gastrointestinal tract in several animal species through secretions, e.g. as lysozime, secretory phospholipase A2 and defensins. The aim of this study was to identify the Paneth cells in the small intestine of baby alpacas. Eighteen culled animals between 1 and 21 days of age were used. Duodenum, jejunum and ileum samples were taken. Samples were fixed in 10% formalin, processed and stained with haematoxylin-eosin (H-E) and Masson trichromic. An anti-lizozime polyclonal antibody was used for immunohistochemistry. Typical Paneth cells were not identified with H-E and Masson trichromic stains, whereas lysozime granules were identified in the base of the crypts of Lieberkühn of duodenum, jejunum and ileum by immunohistochemistry since the first day of age. The number in jejunum and ileum was greater bet...

Reactive oxygen species (ROS) represent a number of highly reactive oxygen-derived by-products generated by the normal mitochondrial respiration and other cellular metabolic reactions. ROS can oxidize macromolecules including lipids, proteins, and nucleic acids. Under physiological condition, the cellular levels of ROS are controlled by several antioxidant enzymes. However, an imbalance between ROS production and detoxification results in oxidative stress, which leads to the accumulation of macromolecular damage and progressive decline in normal physiological functions. Oxidative deterioration of DNA can result in lesion that are mutagenic and contribute to aging and age-related diseases. Therefore, methods for the detection of ROS and oxidative deterioration of macromolecules such as DNA in cells provide important tool in aging research. Here, we described protocols for the detection of...
