Objective: PI3K is one of the most frequently mutated proteins in cancer, resulting in changes to its functions in regulating metabolism, immunity, among others. Despite the identification of specific drugs targeting PI3K, significant resistance tothese therapies has been observed. Therefore, the search for new inhibitors is crucial. This project proposes a strategy based on in silico computational tools for screening Food and Drug Administration (FDA)-approved drugs, aiming to evaluate their potential for drug repositioning. Analiza la actividad de la vía PI3K en células de adenocarcinoma de pulmón ALK+ resistentes a crizotinib, ceritinib y alectinib, con el fin de sugerir posibles nuevos inhibidores utilizando herramientas bioinformáticas para el reposicionamiento farmacológico. A partir de líneas resistentes generadas en laboratorio, se evaluó la actividad de crizotinib, ceritinib y alectinib. La expresión de PI3K y otras proteínas de la vía fue evaluada con western blot. Para el reposicionamiento farmacológico se buscó la secuencia de PI3Ka (p110) en UNIPROT, el modelo farmacofórico y el cribado virtual se realizaron en Pharmit™. Las interacciones entre átomos de inhibidores y aminoácidos de PI3Kα, así como su posición XYZ, se identificaron en Pharmit™ y PLIP™. Competency-based learning in scientific research in the medicine faculties has been the subject of several theoretical approaches and many implementation experiences worldwide. We present the experience of an private university located in the city of Lima, Peru. This university has implemented a strategy of «research-based teaching» in conjunction with «research-oriented teaching» in order to incorporate the achievement of competences for scientific research in the curricular design. The experience is added to those described in medical schools, but with the appropriateness regarding the context of medical education in Peru. The implementation of the curricular strategy has generated unforeseen challenges in the formulation of the intervention, which are described for consideration in new scenarios.
