Polyribosyl-ribitol-phosphate (PRP) from Haemophilus influenzae type b (Hib) is an active immunizing molecule used in the production of the vaccine against H. influenzae, and industrial production could contribute to satisfying a world demand especially in developing countries. In this sense, the aim of this study was to establish a scale-up process using the constant oxygen mass transfer coefficient (kLa) such as the criterion for production of PRP in three different sizes of bioreactor systems. Three different kLa values (24, 52 and 80 h−1) were evaluated in which the biological influence in a 1.5 L bioreactor and 52 h−1 was selected to scale-up the production process until a 75 L pilot-scale bioreactor was achieved. La leucemia linfoblastica aguda (LLA) es una neoplastia común en pediatría, en el Perú se reportan alrededor de 400 casos nuevos al año sólo en infantes. Dentro de los esquemas terapéuticos para tratar la LLA, la enzima L-asparaginasa de origen bacteriano es incluida como biofármaco de primera línea dentro de los principales protocolos de tratamiento. Sin embargo, la L-asparaginasa posee limitaciones que afectan su uso terapéutico y que son producto de su origen procariótico. En ese sentido, la búsqueda de nuevas versiones de la enzima con características cinéticas y estructurales obtenidas de microorganismos poco estudiados y aislados de ambientes inusuales se plantea como una oportunidad con alto potencial para suministrar biofármacos novedosos. Busca optimizar la producción de proteína celular por candida sorboxilosa LBTF.Lev5 mediante aprovechamiento del agua residual procedente de la remoción del mucílago de café (Coffea arabica) para uso potencial como complemento alimenticio de animales. La producción de café es una actividad agroindustrial importante en el Perú. Sin embargo, durante el procesamiento húmedo de los granos de café se generan grandes volúmenes de efluentes de aguas residuales que impactan negativamente en los ecosistemas debido a su descarga inadecuada en cuerpos de agua cercanos. “Aeromonas caviae CHZ306, a marine-derived bacterium isolated from zooplankton, can use chitin (a polymer of a β-(1,4)-linked N-acetyl-D-glucosamine) as a carbon source. The chitin is hydrolyzed by chitinolytic enzymes, namely endochitinases and exochitinases (chitobiosidase and N-acetyl-glucosaminidase). Indeed, the chitinolytic pathway is initiated by the coexpression of the enzymes endochitinase (EnCh) and chitobiosidase (ChB); however, few studies, including biotechnological production of these enzymes, have been reported, although chitosaccharide are helpful in several industries, such as cosmetics. “Microbial proteases, especially aspartic proteases, are an essential group of enzymes produced from different microorganisms. Microbial proteases have several applications, mainly in the food, beverage, cosmetic, and pharmaceutical industries, due to their efficiency in the processing and in the manufacturing stages. The yeast Rhodotorula mucilaginosa CBMAI 1528 was isolated from the Antarctic environment and was previously reported to have higher extracellular aspartic protease production. “Microorganisms from extreme environments, such as the Antarctic ecosystems, have a great potential to produce enzymes with novel characteristics. Within this context, L-asparaginase (ASNase) obtained from yeast species has been poorly studied. In this study, yeasts isolated from samples collected at Admiralty Bay (King George Island, Antarctica) were tested to produce ASNase. From an initial screening of 40 strains, belonging to 13 different species, Leucosporidium scottii L115 produced an ASNase activity (LsASNase activity: 6.24 U g-1 of dry cell weight) with the lowest glutaminase activity. The LsASNase was purified 227-fold, with a specific activity of 137.01 U mg-1 at 37 ◦C, without glutaminase activity.
