The ‘sabaleta’ (Brycon henni) is a species of commercial interest in Colombia. One of the main barriers to the conservation processes of this species is the temporal asynchrony between the reproductive cycles of females and males, which could be solved using biotechnological processes. The aim of this investigation was to evaluate the in vitro maturation of ‘sabaleta’eggs (Brycon henni) using two hormonal preparations. A total of 1200 eggs were subjected to in vitro maturation in the presence of carp pituitary extract (EPC), salmon gonadotropin-releasing hormone (sGnRHa) and a control without hormonal stimulation. The aim of this study was to evaluate three concentrations of centrifuged egg yolk in a commercial diluent for bovine semen. Ejaculates from four bulls were obtained and semen collections were made by electroejaculation and artificial vagina. The sperm obtained were divided into three aliquots supplemented with 10, 20 and 30% centrifuged egg yolk (YHC). Semen freezing was done in 0.5 ml straws. Thawed semen was evaluated for total (MT) and progressive motility (MP), vitality (VE), abnormal morphology (MA) and plasma membrane integrity (MI). Statistical analysis was performed by adjusting generalized linear models (GLM) and comparing means by the Tukey test. La sabaleta (Brycon henni) es una especie de interés comercial en Colombia. Una de las principales barreras para los procesos de conservación de esta especie es la asincronía temporal entre los ciclos reproductivos de las hembras y los machos, que podría ser solucionado mediante el uso de procesos biotecnológicos. El objetivo de esta investigación fue evaluar la maduración in vitro de ovas de sabaleta (Brycon henni) mediante el uso de dos preparaciones hormonales. The aim of this study was to evaluate the cryopreservation of Brycon henni semen using three programmable freezing curves and two permeable cryoprotectants. Semen of 80 males were diluted in medium supplemented with ethylene glycol (EG) or dimethylsulfoxide (DMSO), and cryopreserved in semen straws using three programmable freezing curves: slow (66 min, -0.42 °C/min), medium (43.3 min, -0.6 °C/min) and ultra-rapid (7.7 min, -5.19 °C/min). After one month of storage, semen was thawed and motility was evaluated through a class analysis system (SCA®) and sperm vitality (EV) by fluorescence microscopy with SYBR14/IP probes. The aim of this study was to evaluate the effect of cryopreservation in the presence of sperm subpopulations, according to motility, in sperm ejaculates using the Computer Assisted Semen Analysis (CASA) system. The motility parameters were analysed with principal component analysis (PCA) where they showed the highest variance, thus reducing the number of variables. In the evaluation of 23 738 fresh spermatozoa, the subpopulation (Sp) 1 consisted of progressive spermatozoa and median progressive motility (18.34%), Sp 2 in high velocity and progressive spermatozoa (20.53%), Sp 3 in high-activity spermatozoa, but not progressive (46.79%) and Sp 4 in little activity and non-progressive spermatozoa (14.32%), showing significant differences in the distribution of the four Sp (p<0.001). El objetivo de este estudio fue evaluar el efecto de la criopreservación en la presencia de subpoblaciones de espermatozoides, según la motilidad, en eyaculados de macho cabrío utilizando el sistema de Análisis de Semen Asistido por Computador (CASA). Los parámetros de motilidad se analizaron con análisis de componentes principales (PCA) donde evidenciaron la mayor varianza, reduciendo así el número de variables.
