Sumario:Whit the objective of development an appropriate methodology for induction of direct somatic embryogenesis in Phalaenopsis amabilis (L.) 81., from leaf segments were used immature leaf explants of 15 mm., obtained from flasks that were seeded dehiscent capsule. Leaf segments were evaluated for callus induction, using 12 treatments with different concentrations of auxin-cytokinin, naphthaleneacetic acid (0.00, 0.1 O and 1.00 mg/I) and thidiazurón (0.00, 0.1 O; 1.00 and 3.00 mg/I), accompanied by a basic culture medium consisting of mineral compounds (Murashige y Skoog, 1962) supplemented with 10.00 mg/I Myo-inositol, 0.50 mg/I Pyridoxine, 0.10 mg/I Thiamine, 2.0 mg/I Glycine, 170.00 mg/I NaH2P04, 2.0 g/I peptone, 20.00 g/I sucrose, 3.75 g/I Agar - Agar. Continuing with the process of induction of callus explants were incubated in darkness, grown in a culture medium for a period of 30-45 days in dark conditions and then subject to light, The percentage of callus induction in all treatments evaluated shows that at 30 days of induction there is a 100% reactivity callogénica, the appearance of globular pro-embryonic stages of Phalaenopsis amabilis (L.) BI., demonstrated in treatments T1, T2, Ts y Ts, and the confirmation the tirst embryonic stage (globular stage) show in treatments T5 y Ts, subsequently formed plants. Moreover include that during the development of direct somatic embryogenesis were identified embryonic creamy calli. key words: Pha/aenopsis amabi/is (L.) 81., naphthaleneacetic acid, thidiazuron, explant.
