Departamento Académico de BiologíaLa presente investigación tuvo como objetivo la caracterización del gen responsable de la actividad fructano-exohidrolasas a partir de las raíces reservantes de yacón (Smallanthus sonchifolius) obtenidas de la estación experimental “Baños del Inca” en Cajamarca, perteneciente al Instituto Nacional de Innovación Agraria (INIA). La estandarización de un protocolo de extracción de RNA total a partir de las raíces reservantes de yacón se realizó utilizando el protocolo CTAB, las modificaciones del protocolo se basaron en el aumento de antioxidantes. El diseño de primers permitió la obtención de secuencias codificantes traslapadas del gen FEH mediante la técnica RT-PCR, así como para obtención del extremo 3´ del ARNm. Para esta región se utilizó técnicas variantes de PCR como: RACE, IPCR y semi-Nested, siendo esta última esencial para la obtención del extremo no codificante 3´. El ensamblaje de los fragmentos permitió determinar cerca del 80 por ciento de la secuencia y la construcción del árbol filogenético estableció que la secuencia en estudio corresponde a la 1-fructano exohidrolasa (1-FEH) en yacón, presentando un alto grado de identidad respecto a las 1-FEHs de: C. intybus (88.5 por ciento), H. tuberosus (88.2 por ciento), V. herbácea (86.5 por ciento) y A. lappa (85 por ciento). La propuesta de la estructura de la enzima por homología, realizada mediante los programas: Swiss Model y PyMOL mostró similitud estructural respecto a la estructura base de las glicósido hidrolasa 32, así también la presencia de dos de los tres residuos claves pertenecientes a la triada catalítica y la ausencia de sitios de N-glucosilación cerca a la hendidura.The objective of the present investigation was to characterize the gene responsible for the fructan-exohydrolases activity from the reservant roots of yacon (Smallanthus sonchifolius) obtained from the "Baños del Inca" experimental station in Cajamarca, belonging to the National Institute of Agrarian Innovation (INIA). The standardization of a total RNA extraction protocol from yacon reservant roots was carried out using the CTAB protocol, the modifications of the protocol were based on the increase of antioxidants. The design of primers allowed obtaining of overlapping coding sequences of the FEH gene by the RT-PCR technique, as well as for obtaining the 3'-end of the mRNA. For this region, PCR techniques were used, such as: RACE, IPCR and semi-Nested, the latter being essential for obtaining the 3'-non-coding end. The assembly of the fragments allowed to determine about 80 percent of the sequence and the construction of the phylogenetic tree established that the sequence under study corresponds to 1-fructan exohydrolase (1-FEH) in yacon, presenting a high degree of identity with respect to the 1-FEHs of: C. intybus (88.5 percent), H. tuberosus (88.2 percent), V. herbacea (86.5 percent) and A. lappa (85 percent).
