| Sumario: | Objective: The objective of the present study was to evaluate the antibacterial effect of Peruvian propolis and the action of a methanolic fraction on an in vitro biofilm of Streptococcus gordonii ATCC® 51656™ and Fusobacterium nucleatum ATCC® 10953™. Materials and Methods: Through sequence experiments, 13 propolis from the Peruvian Andes were collected and evaluated by measuring their antibacterial effect on S. gordonii and F. nucleatum strains through inhibition halos. Chlorhexidine and DMSO were used as controls. Organic raw extracts, partitions and propolis fractions were evaluated; besides, it was evaluated propolis cytotoxicity on HGF-1-ATCC CRL-2014 cell lines through colorimetric techniques of 3-(4.5-dimethylthiazole-2-yl) -2.5-diphenyltetrazolium bromide (MTT) and optical density. Finally, the effect of propolis on biofilm was evaluated by confocal microscopy and PCR. Results: It was found that Oxapampa's propolis sample presented antibacterial effect on both strains studied, in raw extracts, chloroformic partitions, butanolic and methanolic fraction. The minimum inhibitory concentration of 0.78 mg/mL of the methanolic fraction of the chloroformic residue of Oxapampa propolis was not toxic on gingival fibroblasts. Concentrations of 0.78 mg/mL and 1,563 mg/mL of the methanolic fraction of the chloroformic residue of propolis from Oxapampa influenced biofilm thickness and copy number of the srtA gene of S. gordonii and radD gene of F. nucleatum at 120 hours, Conclusion: Of the 13 Peruvian propolis, it was found that the methanolic fraction of the chloroformic residue of Oxapampa propolis presented an antibacterial effect on S. gordonii and F. nucleatum and effect on the thickness of the biofilm created in vitro. |
