Using a small group of transcription factors (c-Myc, Oct4, Klf4 and Sox-2), fibroblasts in pluripotent stem cells (iPSC) were converted. This stem cell technology currently has some disadvantages, because there are differences in gene expression profiles due to retention of an "epigenetic memory" from the cells from which they were derived; this could cause problems when applying them in therapy. In this review we discuss the role of some transcription factors involved in iPSCs and ESCs.En las células madre desde que se descubrió su potencial terapéutico se han buscado células madre con capacidad pluripotente, y desde entonces las células madre embrionarias (ESCs) se convirtieron en un centro importante en la terapia celular basada en células madre, ya que a partir de éstas se forman todos los tejidos del cuerpo, pero involucra problemas éticos por el uso de embriones. Posteriormente al usar un pequeño grupo de factores de transcripción (c-Myc, Oct4, Klf4 y Sox-2) se convirtió fibroblastos en células madre pluripotenciales (iPSC). Esta tecnología de células madre actualmente está teniendo ciertas desventajas, debido a que existen diferencias en los perfiles de expresión génica ya que conservan una “memoria epigenética” de las células donde derivan, lo que podría causar problemas al momento de aplicarlas en terapia.
