Gerencia Departamental LoretoEl principal objetivo del presente estudio clínico de Fase l consistió en evaluar la seguridad y tolerancia del extracto acuoso liofilizado del cormo de la planta medicinal Dracontium loretense K. Krause conocido en la Amazonia Peruana como "Jergón sacha". La administración del extracto fue mediante el sistema terapéutico de la microdosis vía intramuscular; el ensayo se definió como de tipo prospectivo, al azar simple, ciego, controlado y unicéntrico; el grupo de estudio estuvo representado por una muestra de 50 voluntarios sanos distribuidos en 3 grupos: 20 recibieron 03 dosis unitarias de 0.025mg, 20 recibieron 0.050mg y 10 recibieron 0.1 ml de agua destilada con un intervalo de 4 días por dosis en cada grupo. La evaluación comprendió análisis de los perfiles bioquímicos y examen médico pre y post tratamiento. Los resultados demuestran que las dosis administradas no ocasionan efectos adversos, asimismo no alteran los signos vitales, revelando de esta manera seguridad y tolerancia; reportándose paralelamente resistencia al esfuerzo y ganancia de peso, mejorando consecuentemente el estado general de los voluntarios, apreciándose además en la evaluación linfocitaria una acción inmunomoduladora, corroborado por las pruebas pre-clínicas de inmunoestimulación efectuadas con ratas. A la dosis de 0.050mg se presentó somnolencia y cefalea pasajera con aumento de apetito como efectos secundarios en un bajo porcentaje de voluntarios, manifestándose también en el grupo control. Las evidencias encontradas hacen permisible la aplicación sin mayor riesgo del extracto de esta especie vegetal en estudios clínicos de Fase II para su evaluación en patologías específicas.The primary objective of the present clinical study of Phase I consists in evaluating the safety and tolerance of the watery, liophilizated extract of the cormo of the medicinal plant Dracontium loretense K. Krause known in the Peruvian Amazonian as "Jergón Sacha". The administration of the extract was through an intramuscular therapeutic system of the microdose; the test was defined by the prospective type at random, blind, and centrally controlled; the study group was represented by 50 healthy volunteers divided into three groups: 20 received 3-unit doses of 0.025mg, 20 received 0.050mg and 10 received 0.1 ml of distil water with an interval of four days between each dose in each group. The evaluation comprised an analysis of the biochemical profiles and a pre- and post-treatment medical exam. The results demonstrate that the administered doses do not cause adverse effects, nor do they alter vital signs, revealing safety and tolerance, parallelly reporting strength resistance and weight gain, consequently, improving the general state of the volunteers, appreciated in the limphocitarie evaluation an inmunemoduler action, the evidence corroborates the pre-clinical immunostimulation encountered in mice. The dose of 0.050mg presented drowsiness and passing fleeting migraine with an increase in appetite as secondary effects in a low percentage of volunteers, manifested as well in the. control group.
