The present study evaluated the effect of temperature (12-15 and 22-25 °C) and the time of storage (0 and 16 hours) in ovaries of alpacas on the maturation and cleavage rate post in vitro fertilization. The ovaries were collected from slaughtered mature alpacas at Pilpichaca slaughterhouse (Huancavelica, Peru) and transported to the laboratory in 0.9% saline solution with an initial temperature of 35 °C. The ovaries were randomly distributed in three groups: Control (0 h), T1 (16 h at 12-15 °C) and T2 (16 h at 22-25 °C). The oocytes were recovered by slicing of the ovaries and only oocytes with one or more layers of cumulus were selected and cultured in maturation medium TCM-199 for 40 h under 5% CO2 at 39 °C. Se evaluó el efecto de la temperatura (12-15 y 22-25 ºC) y el tiempo de almacenamiento (0 y 16 horas) de ovarios de alpacas sobre la tasa de maduración y división pos-fecundación in vitro. Los ovarios fueron recolectados de alpacas adultas sacrificadas en el camal de Pilpichaca (Huancavelica, Perú) y transportados al laboratorio en solución salina al 0.9% con una temperatura inicial de 35 ºC. Los ovarios fueron distribuidos al azar en los tratamientos Control (0 h), T1 (16 h a 12-15 ºC) y T2 (16 h a 22-25 ºC). Los ovocitos se obtuvieron por disección, seleccionándose aquellos con una o más capas de cúmulos, y madurados en medio TCM 199 por 40 h bajo condiciones de 5% CO2 y 39 °C. The study aimed to evaluate the effect of three cryoprotectants: Dimetil Sulfoxide (DMSO), Ethylene glycol (EG) and Glycerol (GL) on the cryopreservation of epididymal alpaca sperm. Testicles of alpacas older than 3 years were used. The epididymides were separated and the tails were dissected. The spermatozoa were recovered with fraction A of the extender (cow skim milk, egg yolk and fructose), selecting samples with sperm motility equal or higher than 50% (n=18, motility = 69%, sperm membrane functional integrity = 48%). The fraction A with the sperm was refrigerated (cooling rate of 1 ºC/5 min) until 5 ºC. The fraction was divided in three and added the same volume of fraction B with the cryoprotectant (DMSO: 7%, 0.9M; EG: 7%, 0.9M; GL: 7%, 1.2M).
