The funders had no role in study design, data collection and analysis, decision to publish, or preparation of the manuscript.Racemose neurocysticercosis is an aggressive disease caused by the aberrant expansion of the cyst form of Taenia solium within the subarachnoid spaces of the human brain and spinal cord resulting in a mass effect and chronic inflammation. Although expansion is likely caused by the proliferation and growth of the parasite bladder wall, there is little direct evidence of the mechanisms that underlie these processes. Since the development and growth of cysts in related cestodes involves totipotential germinative cells, we hypothesized that the expansive growth of the racemose larvae is organized and maintained by germinative cells. Here, we identified proliferative cells expressing the serine/threonine-protein kinase plk1 by in situ hybridization. Proliferative cells were present within the bladder wall of racemose form and absent from the homologous tissue surrounding the vesicular form. Cyst proliferation in the related model species Taenia crassiceps (ORF strain) occurs normally by bud-ding from the cyst bladder wall and proliferative cells were concentrated within the growth buds. Cells isolated from bladder wall of racemose larvae were established in primary cell culture and insulin stimulated their proliferation in a dose-dependent manner. These findings indicate that the growth of racemose larvae is likely due to abnormal cell proliferation. The different distribution of proliferative cells in the racemose larvae and their sensitivity to insulin may reflect significant changes at the cellular and molecular levels involved in their tumor-like growth. Parasite cell cultures offer a powerful tool to characterize the nature and formation of the racemose form, understand the developmental biology of T. solium, and to identify new effective drugs for treatment. © 2021 Orrego et al.Fondo Nacional de Desarrollo Científico y Tecnológico - FondecytengPublic Library of SciencePLoS Neglected Tropical Diseasesinfo:eu-repo/semantics/openAccesshttps://creativecommons.org/licenses/by/4.0/protein serine threonine kinaseBeta actin-1complementary DNA-1insulin-1polo like kinase 1-1parasite antigen-1http://purl.org/pe-repo/ocde/ford#4.04.01-1Identification and culture of proliferative cells in abnormal taenia solium larvae: Role in the development of racemose neurocysticercosisinfo:eu-repo/semantics/articlereponame:CONCYTEC-Institucionalinstname:Consejo Nacional de Ciencia Tecnología e Innovacióninstacron:CONCYTEC#PLACEHOLDER_PARENT_METADATA_VALUE##PLACEHOLDER_PARENT_METADATA_VALUE##PLACEHOLDER_PARENT_METADATA_VALUE##PLACEHOLDER_PARENT_METADATA_VALUE##PLACEHOLDER_PARENT_METADATA_VALUE##PLACEHOLDER_PARENT_METADATA_VALUE##PLACEHOLDER_PARENT_METADATA_VALUE#ORIGINALIdentification and culture - journal.pntd.0009303.pdfIdentification and culture - journal.pntd.0009303.pdfapplication/pdf3498728https://repositorio.concytec.gob.pe/bitstreams/d1e14973-92fe-4c41-9925-7faee962fe1e/download52f50677e4eccfa0747e6234369275d6MD51TEXTIdentification and culture - journal.pntd.0009303.pdf.txtIdentification and culture - journal.pntd.0009303.pdf.txtExtracted texttext/plain60408https://repositorio.concytec.gob.pe/bitstreams/38fadd46-98c9-49a5-bcc1-b9d0f567e267/downloade4020548ccb899d5096a1d98a04c5b62MD52THUMBNAILIdentification and culture - journal.pntd.0009303.pdf.jpgIdentification and culture - journal.pntd.0009303.pdf.jpgGenerated Thumbnailimage/jpeg5835https://repositorio.concytec.gob.pe/bitstreams/1bd916a3-bd72-4d65-bcc2-808b95b2b718/downloadc24a3f42fad1bcef593b256b0e213bc6MD5320.500.12390/2376oai:repositorio.concytec.gob.pe:20.500.12390/23762025-01-19 22:00:23.463https://creativecommons.org/licenses/by/4.0/info:eu-repo/semantics/openAccesshttp://purl.org/coar/access_right/c_abf2info:eu-repo/semantics/openAccessopen accesshttps://repositorio.concytec.gob.peRepositorio Institucional CONCYTECrepositorio@concytec.gob.pe#PLACEHOLDER_PARENT_METADATA_VALUE##PLACEHOLDER_PARENT_METADATA_VALUE##PLACEHOLDER_PARENT_METADATA_VALUE##PLACEHOLDER_PARENT_METADATA_VALUE##PLACEHOLDER_PARENT_METADATA_VALUE##PLACEHOLDER_PARENT_METADATA_VALUE##PLACEHOLDER_PARENT_METADATA_VALUE#<Publication xmlns="https://www.openaire.eu/cerif-profile/1.1/" id="3eb3acba-bc72-47a1-ad1b-dde8b9165dfc"> <Type xmlns="https://www.openaire.eu/cerif-profile/vocab/COAR_Publication_Types">http://purl.org/coar/resource_type/c_1843</Type> <Language>eng</Language> <Title>Identification and culture of proliferative cells in abnormal taenia solium larvae: Role in the development of racemose neurocysticercosis</Title> <PublishedIn> <Publication> <Title>PLoS Neglected Tropical Diseases</Title> </Publication> </PublishedIn> <PublicationDate>2021</PublicationDate> <DOI>https://doi.org/10.1371/journal.pntd.0009303</DOI> <SCP-Number>2-s2.0-85103804478</SCP-Number> <Authors> <Author> <DisplayName>Orrego, Miguel A.</DisplayName> <Person id="rp05774" /> <Affiliation> <OrgUnit> </OrgUnit> </Affiliation> </Author> <Author> <DisplayName>Verastegui, Manuela R.</DisplayName> <Person id="rp05775" /> <Affiliation> <OrgUnit> </OrgUnit> </Affiliation> </Author> <Author> <DisplayName>Vasquez, Carlos M.</DisplayName> <Person id="rp05772" /> <Affiliation> <OrgUnit> </OrgUnit> </Affiliation> </Author> <Author> <DisplayName>Koziol, Uriel</DisplayName> <Person id="rp05776" /> <Affiliation> <OrgUnit> </OrgUnit> </Affiliation> </Author> <Author> <DisplayName>Laclette, Juan P.</DisplayName> <Person id="rp05773" /> <Affiliation> <OrgUnit> </OrgUnit> </Affiliation> </Author> <Author> <DisplayName>Garcia, Hector H.</DisplayName> <Person id="rp00672" /> <Affiliation> <OrgUnit> </OrgUnit> </Affiliation> </Author> <Author> <DisplayName>Nash, Theodore E.</DisplayName> <Person id="rp00750" /> <Affiliation> <OrgUnit> </OrgUnit> </Affiliation> </Author> </Authors> <Editors> </Editors> <Publishers> <Publisher> <DisplayName>Public Library of Science</DisplayName> <OrgUnit /> </Publisher> </Publishers> <License>https://creativecommons.org/licenses/by/4.0/</License> <Keyword>protein serine threonine kinase</Keyword> <Keyword>Beta actin</Keyword> <Keyword>complementary DNA</Keyword> <Keyword>insulin</Keyword> <Keyword>polo like kinase 1</Keyword> <Keyword>parasite antigen</Keyword>
Racemose neurocysticercosis is an aggressive disease caused by the aberrant expansion of the cyst form of Taenia solium within the subarachnoid spaces of the human brain and spinal cord resulting in a mass effect and chronic inflammation.
