Maestría en Producción AnimalLa incorporación de ciclodextrinas cargadas de colesterol (CLC), representa una estrategia para aumentar la resistencia de espermatozoides al proceso de criopreservación, mejorando las tasas de supervivencia espermática y la calidad del semen después de la descongelación con respecto al no tratado. El objetivo de esta investigación fue determinar el efecto de la adición de ciclodextrina cargada con colesterol sobre la criopreservación de semen de bovino en la raza Holstein Friesian. El estudio se llevó a cabo en el Laboratorio del Banco Nacional de Semen de la UNALM, utilizando 4 toros Holstein Friesian de 3 a 4 años de edad. Se colecto utilizando vagina artificial un total de 28 eyaculados (7 eyaculados por toro) y se analizó sus características macroscópicas (color, volumen, pH) y microscópicas (motilidad, vitalidad, concentración, integridad y funcionalidad de la membrana espermática). Posteriormente se realizó una pre dilución 1:1 del semen y se dividió en 03 alícuotas para la adición de CLC (T1: 0 mg, T2:1.5 mg. CLC y T3:2 mg de CLC /120 millones de espermatozoides). Se incubo por 15 minutos a 37ºC; luego se realizó la dilución final y el inmediato envasado del semen en pajillas 0,5 ml (30x106 espermatozoides) realizando el descenso escalonado de temperatura, llegando a -196º C. Se almaceno en tanques de nitrógeno líquido; a los 7 días se procedió a descongelar las pajillas para su evaluación. Se empleó el DBCA con sub muestreos para medir el efecto de la CLC en los diferentes tratamientos. Se determinó que no existió diferencia significativa en la etapa de refrigeración en los diferentes tratamientos. Resultados post-descongelamiento indican una mejora en las diferentes variables estudiadas utilizando CLC, la motilidad total espermática fue de 67.73%, 79.59% y 74.73% con T1, T2 y T3 respectivamente. Vitalidad con T1: 63.26%, T2:75.84% y T3:70.53%. Valores a prueba de HOST en T1, T2 y T3 con el 46.89%, 60.43% y 55.06% respectivamente.The incorporation of cholesterol-loaded cyclodextrins (CLC) represents a strategy to increase the sperm resistance to the cryopreservation process, improving the sperm survival rates and the quality of the semen after thawing with respect to the untreated one.
