To evaluate the oncolytic activity of the rLS1 viral construct on prostate carcinoma (DU-145) and renal cancer (786-O) cell lines, as well as its selective replication in both tumor lines and the non-tumoral madin-darby canine kidney (MDCK) cell line. Methods. The oncolytic effectiveness was determined by reducing cell viability and apoptosis using the (3-[4,5-dimethylthiazol-2-yl]-5-[3-carboxymethoxyphenyl]-2-[4-sulfophenyl]-2h-tetrazolium) MTS colorimetric assay and by labeling with annexin v/anxa5-pe/sytox blue dead cell, respectively. The viral replicative selectivity in cells was evaluated by crystal violet staining. Results. The MTS assay determined that rls1 significantly reduced viability (p < 0,001) in both tumor cell lines without causing a significant reduction in the viability of MDCK cells compared to uninfected cells. A significant increase (p < 0,001) in the percentage of apoptotic cells was observed in 786-O and DU-145 cells infected with rLS1 compared to uninfected cells. Conclusion. The results indicate that rLS1 demonstrated selective oncolytic potential in human prostate and renal cancer cells, suggesting its potential as an antitumoral agent. |

La viroterapia oncolítica se ha convertido en un método novedoso para el tratamiento del cáncer. El virus de la enfermedad de Newcastle (VENC) presenta potencial oncolítico y replicación selectiva en células tumorales. El cáncer de próstata y renal humano son una de las principales causas de muerte a nivel mundial. Objetivo. Evaluar la actividad oncolítica del constructo viral rLS1 sobre las líneas celulares de carcinoma prostático (DU-145) y cáncer renal (786-O), y evaluar su replicación selectiva en ambas líneas tumorales y en la línea celular no tumoral de riñón canino madin-darby (MDCK). Métodos. La efectividad oncolítica fue determinada mediante la reducción de viabilidad celular y apoptosis, usando el ensayo colorimétrico (3-[4,5-dimetiltiazol-2-il]-5-[3-carboximetoxi-fenil]-2-[4-sulfofenil]-2h-tetrazolio) (MTS) y el marcaje con annexin v/anxa5-pe/sytox blue dead cell, respectivamente. La selectividad replicativa viral en las células fue evaluada por tinción con cristal violeta. Resultados. El ensayo de MTS determinó que el virus rLS1 redujo significativamente el porcentaje de viabilidad celular (p < 0,001) en ambas líneas celulares tumorales, sin causar una reducción significativa de la viabilidad celular en las células MDCK en comparación a las células no infectadas. Se demostró un aumento significativo (p < 0,001) del porcentaje de células apoptóticas en las células 786-O y DU-145 infectadas con rls1 en comparación a las células no infectadas.
